Factor B is a serine protease zymogen of molecular weight 93 kDa. Usually occurring in serum 150-260 mg/L. It is the central enzyme of the alternative pathway of complement activation. There appears to be a natural and continual low level activation of C3 occurring in the body. This can lead to the deposition of C3b on a bacterial surface where it acts as a substrate for factor B, to produce C3bB. Factor D, a low level serum protein, is then able to cleave bound factor B to produce C3bBb with the peptide Ba being released to the fluid phase. Another serum protein, factor P (properdin) then stabilizes the C3bBb complex. This is the alternate pathway convertase, which can cleave further C3molecules to C3a and C3b (amplification loop) or cleave C5 leading into the common terminal sequence activation and cell lysis.
