Changes in chromatin structure play a large role in the regulation of transcription in eukaryotes . The nucleosome is the primary building block of chromatin, and is made up of four core histone proteins (H2A, H2B, H3 and H4) . Various post-translational modifications, such as acetylation and methylation, of core histones serve as regulators of gene expression . Histone H3 is primarily acetylated at lysines 9, 14, 18, and 23 . Methylation also serves as a predominant form of post-translational modification of Histone H3, and has been linked to transcriptional regulation and epigenetic silencing via heterochromatin assembly . Lysines 4, 9 and 27 in the N-terminal tail of Histone H3 are the preferred sites of methylation.
