A member of Pyruvate dehydrogenase kinase (PDK1, 2, 3, and 4), PDK2 is a serine kinase which is involved in the regulation of mammalian glucose metabolism. During aerobic energy metabolism, mitochondrial pyruvate dehydrogenase complex (PDC) catalyzes decarboxylation of pyruvate to form acetyl-CoA . PDK2 inhibits PDC thru phosphorylation of the E1 alpha subunit of PDC and inactivates the complex . Highly expressed in heart and skeletal muscles, PDK2 is also responsible for the phosphorylation of Akt at Ser473, which leads to phosphorylation by PDK1 on Thr308 on Akt and full Akt activation .
