The bicarbonate transporter (BT) superfamily is made up of four known members of the Na+-dependent HCO3- cotransporter isoform family and (NBC1-4) and members of the anion exchanger (AE) family which includes two NBC-related proteins, anion exchanger 4 (AE4) and the Na+-dependent chloride-bicarbonate exchanger (NCBE). Together, these proteins are responsible for the critical regulation of biological fluid and bodily pH via the bicarbonate/CO2 buffering system in epithelial and nonepithelial cells. NBC proteins are broadly expressed and can be found in kidney, pancreas, stomach, large and small intestines, brain, heart, and epididymis. The electrogenic NBC1 protein is expressed in kidney proximal tubule and pancreatic duct cells and plays a major role in bicarbonate reabsorption. NBC1 can be activated by carbonic anhydrase II (CAII) and the cystic fibrosis transmembrane conductance regulator (CFTR) in the pancreas where it effects HCO3- secretion.
